    采用稀释涂布法从发病烟株的根际土壤中分离、纯化得到生防菌LS06,通过平皿对峙培养、16S rRNA测序比对、基因组测序分析和盆栽试验,评价LS06的生防应用潜力。结果表明:LS06对7种镰刀菌均具有抑制作用,抑菌率为36.69%~49.30%;经鉴定,该菌株为短短芽孢杆菌,基因组大小为6 131 499 bp;COG功能注释显示其有5 768个功能基因,其中34个基因与趋化、移动和铁载体合成相关;基因组中有14个次生代谢产物的生物合成基因簇,其中4个基因簇与已知化合物的生物合成基因簇的相似性高于78%,其余10个基因簇的功能产物尚未明确,说明LS06具有合成多种抑菌活性物质的潜力。盆栽试验结果表明LS06对7种镰刀菌的相对防效为64.74%~76.74%。综上所述,LS06可抑制7种主要镰刀菌的生长,具有分泌多种抑菌活性物质的潜力,在镰刀菌根腐病的生物防控中具有良好的应用前景。

    Abstract:

    A biocontrol strain designated LS06 was isolated and purified from the rhizosphere soil of diseased tobacco plants using the dilution plating method. The biocontrol potential of strain LS06 was evaluated through dual culture assays, 16S rRNA gene sequencing and alignment, genome sequencing analysis, and pot experiments. Dual culture assays showed that strain LS06 inhibited seven Fusarium species, with inhibition rates ranging from 36.69% to 49.30%. The strain LS06 was identified as Brevibacillus brevis, with a genome size of 6 131 499 bp. COG annotation indicated that it harbors 5 768 genes, of which 34 genes are associated with chemotaxis, motility, and siderophore synthesis. The genome contains 14 biosynthetic gene clusters(BGCs) for secondary metabolites. Four of these clusters exhibit more than 78% similarity to known BGCs, whereas the products of the remaining 10 clusters remain unknown, suggesting that strain LS06 possesses the potential to synthesize diverse antimicrobial compounds. Pot experiments demonstrated that the relative control efficacy of strain LS06 against the seven Fusarium species ranged from 64.74% to 76.74%. In conclusion, strain LS06 is capable of inhibiting seven major Fusarium species, has the potential to produce various antimicrobial substances, and shows considerable promise for application in the biological control of Fusarium root rot.
