In order to clarify the relationship between the BnaYUCCA10 gene expression and the oleic acid content in Brassica napus, we used miRNA sequencing and real-time fluorescent quantitative PCR(RT-PCR) methods in this study to analyze the expression of BnaYUCCA10 gene in 30 samples collected from different tissues and growth periods of B. napus with different oleic acid content. And whether the oleic acid is involved in the metabolism was also analyzed. The results showed that the expression level of BnaYUCCA10 gene increased gradually with the growth process in most B. napus materials during the vegetative growth stage, and there was no significant difference between high oleic acid materials and non-high oleic acid materials. The expression level of 1 DAF flowers was higher than that of buds and 5 DAF flowers, and the expression level of high oleic acid materials and non-high oleic acid materials was significantly different. The expression level in silique stage decreased gradually with the growth process, and the expression level of high oleic acid materials and non-high oleic acid materials was significantly different. The expression level of the BnaYUCCA10 gene in silique stage and 5 DAF flowers was significantly related to the oleic acid content. The expression level of the high oleic acid materials was significantly higher than that in non-high oleic acid materials. It was suggested that BnaYUCCA10 gene might be involved in oleic acid metabolism in seeds, which could be of insight to the further research on the regulation mechanism of oleic acid in rapeseed.
