磁性氧化石墨烯的制备及其对沙门氏菌荧光探针的猝灭机理

    摘要:

    为探讨磁性氧化石墨烯(Fe3O4/GO)对荧光的猝灭作用,通过扫描电镜(SEM)、透射电镜(TEM)、X射线衍射(XRD)及傅里叶变换红外光谱(FT–IR)对化学共沉淀法制备的Fe3O4/GO和氧化石墨烯(GO)的结构进行对比表征,发现Fe3O4磁性纳米粒子负载在GO的表面,表明成功合成了Fe3O4/GO。以Fe3O4/GO为荧光猝灭剂,沙门氏菌荧光探针为荧光剂,通过荧光、紫外吸收光谱,结合Stern–Volmer方程及Lineweaver–Burk双倒数曲线方程,对其荧光猝灭机理进行初步分析,结果表明:Fe3O4/GO具有荧光猝灭作用,当Fe3O4/GO质量浓度为3.2×10?4 g/mL时,荧光猝灭率可达96.86%,其猝灭为静态猝灭;用Fe3O4/GO荧光探针复合物检测沙门氏菌DNA具有较好的选择性。

    Abstract:

    In order to investigate the quenching effect of magnetic graphene oxide(Fe3O4/GO) on fluorescence, Fe3O4/GO was prepared using chemical method and then characterized using scanning electron microscopy(SEM), transmission electron microscopy(TEM), X-ray diffraction(XRD) and Fourier transform infrared spectroscopy(FT–IR). The results showed that Fe3O4 magnetic nanoparticles successfully synthesized and were loaded on the surface of GO. In its application on quenching, the Fe3O4/GO was used as the fluorescence quenching agent, and the fluorescence probe of salmonella was used as the fluorescence agent. The fluorescence quenching mechanism of Fe3O4/GO was preliminarily analyzed by fluorescence and ultraviolet absorption spectra, combined with Stern-Volmer equation and Lineweaver-Burk double reciprocal curve equation. The results showed that Fe3O4/GO had fluorescence quenching effect. When the concentration of Fe3O4/GO was 3.2×10?4 g/mL, the fluorescence quenching rate was up to 96.86%, in a static quenching way. Moreover, Fe3O4/GO fluorescence probe complex has good selectivity in the detection of salmonella DNA.
