Abstract:The promoter of OsFAH gene was cloned from Oryza sativa cultivar ‘Xiangwanxian 13’, and analyzed for the presence of putative cis-elements. The OsFAH promoter contained basal promoter elements, TATA-box, CAAT-box, and the cis-elements involved in light response. The promoter was cloned into a vector that contains the GUS gene, and introduced into Arabidopsis. GUS expression in the transgenic Arabidopsis was detected by histochemical staining. Strong GUS expression was observed in young cotyledons, hypocotyls, and true leaves. However, low GUS expression was only detected in old leaves. GUS expression was found in the vascular tissues of the taproots. And, GUS expression driven by OsFAH promoter and OsFAH expression were decreased when plants were exposed to darkness.
