Abstract:In this study, we investigated the variables of purification process of polyphenolic compounds from pericarpium trichosanthis and these variable effects on the antioxidation activity of the purified products. The method to purify was macroporous adsorption resin. The results showed that AB-8 macroporous resin has the best adsorption and desorption performance. The optimized adsorption process variables should be at: the concentration of polyphenols of extract 0.6 mg/mL; pH 3.0, the flow rate of fluid 2 BV/h, the volume 4 BV. The optimized elution parameters included the eluting regent 60 % ethanol, eluent volume 6 BV, elution rate 2 BV/h. By use of the optimized process, the purified polyphenol content was 49.52%, 57 times higher than those purified from the conventional process. The antioxidant activity of purification product increased 4-5 times than those form the conventional method. The IC50 of DPPH+ free radical scavenging effect of the polyphenols in pericarpium trichosanthis before and after purification were 2.01 mg/mL and 0.53 mg/mL, respectively, while the IC50 of free radical scavenging effect on ABTS+ was 0.62 mg/mL and 0.13 mg/mL, respectively.
