Abstract:To explore the insecticidal activity and biological characteristics of Tolypocladium inflatum strain SCKDC10, indoor virulence assay was conducted to study the virulence of strain SCKDC10 on 4th instar larvae and pupae of Bombyx mori and on Panonychus citri(McGregor) adult females. The effects of different factors, including culture media (PDA, PSA, SPA, SYA, Czapek’s, SDAY, NA, LB), carbon sources (starch, mannite, inositol, maltose, glucose, lactose, glycerol, sucrose), nitrogen sources (potassium nitrate, urea, peptone, ammonium nitrate, glycine, lysine, ammonium chloride), temperature (10, 15, 20, 25, 28, 30, 35, 40 ℃), pH(4.0, 5.0, 6.0, 7.0, 8.0, 9.0, 10.0), UV irradiation (0, 20, 40, 60, 80, 100, 120 min) and common insecticides(spirodiclofen, fenpropathrin, fenfluidate, clofentezine, thiadenone) and fungicides (tebuconazol, pyraclostrobin, prochloraz, carbendazim, zoledronic acid) on the hyphal growth, sporulation and spore germination of strain SCKDC10 were tested by single factor experiment. The results indicated that the LC50 of strain SCKDC10 to the larvae and pupa of Bombyx mori were 1.47×108 spores/mL and 4.25×108 spores/mL respectively, and the LT50 were 6.51 d and 7.87 d, respectively. The LC50 of the strain to the Panonychus citri(McGregor) was 9.26×107 spores/mL, and the LT50 was 6.34 d. SDAY and PDA are the optimal media for growth and sporulation, respectively. Glucose and glycine are more conducive to the growth and sporulation of the strain. The optimum growth and sporulation temperature for the strain were 28 ℃. And the optimum pH for growth and sporulation of the strain were 7.0. With the prolongation of UV irradiation, the growth rate, sporulation and spore germination rate of the strain decreased gradually. After UV irradiation for 100 min, spore germination was completely inhibited. The duration of UV irradiation had a great influence on the spore germination. Strain SCKDC10 had good compatibility with insecticides but sensitive to fungicides.
