Abstract:To study the optimal condition for the cultivation of Microsporum gypseum isolated from giant panda, different conditions including media, carbon sources, nitrogen sources, temperature and pH were selected as indicators to culture the strain, and the biological characteristics such as the color, morpholo and diameter of colony were observed using cross method every day. The results showed that the optimum medium for M. gypseum growth was SDA(sabouraud dextrose agar), it could also grow well in medium of M–SDA(modify sabouraud dextrose agar), PDA(potato dextrose agar) and TM(takashio mediun), while it grew slowly in medium of DTM(dermatophytes identification Medium) and AA(ascospore agar). Impacts of mediums from five kinds of carbon sources on the growth of M. gypseum have little difference, and among of them, the glucose was the great one. Apart from urea, M. gypseum could grow well in other four nitrogen–containing mediums and peptone was the optimum one. The optimum cultivation temperature was 25 ℃, and M. gypseum could grow well in the temperature ranging from 15 to 35 ℃, but its growth was the worst in 35 ℃.The optimum pH for its growth was 10, M. gypseum grew better in pH 5, 6, 8 and 9 than in pH 7, but when the pH value was in 4 and 11, the worst growth happened.
