Abstract:Enrichment medium added with copper ions was used to screen bacteria possessing laccase activity from birch forest soil. A bacterium with high laccase activity was identified as Bacillus sp. 9BS by morphological, physiological and biochemical characteristics with 16S rDNA sequence analysis. The optimum growth temperature of strain 9BS was 37 ℃ and the optimum growth pH was 7.0. The strain 9BS had the resistance to salt and grew well in 7% NaCl solution. Using syringaldazine as substrate, the laccase activity of the dry spore of Bacillus sp. 9BS was 47.1 U/g. In the absence of mediator, the spore laccase of strain 9BS showed effective decolorization effect on Isatin (IC) and crystal violet (CV), the decolorization rates in 6 h were 73.3% and 82.2%, respectively.
