Abstract:The suitable condition of seeds germination and their growth were researched by taken fresh seeds of Camptotheca acuminate on that very year as materials through plant tissue culture. To compare the rate of seed germination, explants were disinfected with 0.1% HgCl2 and 10% NaClO respectively. The result showed that rate of seed germination was the highest with 0.1% HgCl2 disinfection, which achieved 41.11%. Further, rate of seed germination and seedling were significantly increased with 10% NaClO soak for 3 minutes on the basis of treatment of 0.1% HgCl2 disinfection, which reached 85.56% and 55.56% respectively. Fresh seedlings grew well with an average of 6–7 lateral roots and a 10 cm long taproot. The growth of seedlings treated by root-cut and transferred in fresh MS medium were same as treatment with 10% NaClO. Seedlings, however, became yellow and had no new root or only one short root after transferred them to MS medium with 1 mg/L 6–BA .
