Abstract:Using a pair of degenerate primers designed based on HMG–box,a conserved region in Sox gene, two 217 bp-fragments were amplified and cloned from the male and the female Andrias davidianus which exhibited no sexual difference by PCR–SSCP analysis. Amino acid sequence analysis showed one of the 217 bp-fragments showed 90% homology to Sox11 gene of human, zebrafish, mouse, chicken, Xenopus tropicalis and Alligator sinensis respectively and the other showed 90% homology to Sox14 gene of Tilapia, Ornithorhynchus anatinus, mouse, Fugu rubripes, zebrafish, chicken, human and Xenopus Laeuis respectively, thereafter the two fragments were termed as adSox11 and adSox14c respectively. Sox gene transcripts were found in spermary, ovary, heart and kidney by RT–PCR but not stomach and liver of Andrias davidianus. These results collectively suggested that Sox gene was highly conserved across species Sox gene and was selectively expressed among tissues and organs to accommodate the need of development.
