Abstract:Abstract:In order to optimize electrical activation and oocytes culture condition, the effect of different electrical activation parameters and culture media on parthenogenetic activation of porcine oocytes was investigated. The result indicated that one-time pulse electrical activation under pulse voltage of 150 V/mm and pulse duration of 100 µs was the best condition for parthenogenesis of porcine oocytes. The blastocyst rate in the group treated with a pulse voltage of 150 V/mm reached up to 46.3%, which was significantly higher than that in the groups treated with pulse voltages of 100 V/mm and 125 V/mm respectively under the same condition of pulse duration and pulse times (P<0.05). And the blastocyst rate in the group treated with pulse duration of 100 µs reached up to 50.5%, which was significantly higher than that in the other two groups treated with pulse duration of 75 µs and 125 µs respectively under the same condition of pulse voltage and pulse times (P<0.05). The cleavage rate and the blastocyst rate in the group treated with one-time pulse reached up to 82.7% and 50.3% respectively, which was significantly higher than that in the group treated with two-time pulse under the same condition of pulse voltage and pulse duration (P<0.05). The activated parthenogenetical development of porcine oocytes in groups cultured in media PZM3 and NCSU23 respectively showed no significant difference (P>0.05), but cleavage rate, blastocyst rate and blastocyst cell number in the group cultured in medium PZM3 were a little higher than that in the group cultured in medium NCSU23.
