    运用PCR技术扩增茶尺蠖核型多角体病毒安徽分离株(EcobNPV-AH)和湖北分离株(EcobNPV-HB)的同源重复序列2(hr2),并对湖北分离株的hr2进行克隆和序列测定.结果表明,茶尺蠖核型多角体病毒安徽分离株hr2扩增产物为2条DNA片段,大小约1 300 bp和1 800 bp,湖北分离株的hr2扩增产物为1条DNA片段,大小约1 800 bp,由基因型组成判断,EcobNPV-HB是新的地区分离株,其hr2全长1 761个核苷酸,GC含量21.7%,编码540个氨基酸,具有重复性回文序列,较GenBank中登录的EcobNPV-AH的hr2多362个核苷酸.

    Abstract:

    Homologous repeat region 2 (hr2) of Ectropis obliqua nucleopolyhedrovirus (EcobNPV) isolated from Hubei and Anhui strains were amplified by PCR. The results showed that only one 1.8 kb DNA fragment was amplified from EcobNPV-HB strains and two DNA fragments was amplified from EcobNPV-AH strains, which suggested that EcobNPV-HB strains was an independent strains different from EcobNPV-AH strains. hr2 of Ectropis obliqua nucleopolyhedrovirus isolated from Hubei strains was cloned into pMD18-T vector, sequenced and analyzed. The hr2 of EcobNPV-HB strains was composed of 1 761 nucleotides and coded 540 amino acids with a G+C content of 21.7%, comprised of perfect palindromes. Extra 362 nucleotides were observed compared with hr2 of EcobNPV-AH strains in GenBank.
