多效唑对大蒜试管苗鳞茎化培养的影响

    摘要:

    以MS为基本培养基,用大蒜茎尖外植体培养的试管苗进行离体培养试验。结果表明,一定范围内,随着多效唑浓度增加,抑制鳞茎形成与分化,叶片和根系变短增粗,添加6mg?L-1多效唑为诱导鳞茎形成和促进膨大的最佳浓度;内源激素含量变化表明,多效唑处理后,大蒜试管苗叶鞘中IAA/ABA的数值急剧下降,降低幅度越大,鳞茎形成膨大效果越好。

    Abstract:

    In vitro experiments were carried out,regarded test-tube seedings of garlic stem tip as explants and took MS as the basic culture medium.The results showed that Within a certain range,raising concentration could inhibit the formation and differentiation of the bulb, the leaves and roots were shortened and widened. The 6mg?L-1 PP333 was the optimal treatment concentration of the inducing bulb formation and development. The endogene hormone content change showed that after PP333 treatment, in the garlic test tube seedlings leaf sheath the IAA/ABA ratio drops suddenly,and reduces the scope to be bigger, the bulb forms expands the effect to be better.
