      将30只SD大鼠按体质量随机分为正常组,模型组,L–茶氨酸低剂量(100 mg/kg)、中剂量(200 mg/kg)、高剂量(400 mg/kg)组。采用每日皮下注射200 mg/kg D–半乳糖生理盐水溶液的方式建立衰老大鼠模型,考察L–茶氨酸对D–半乳糖致衰老大鼠肝脏谷丙转氨酶(ALT)和谷草转氨酶(AST)活性、晚期糖基化终末产物(AGEs)和炎性介质含量的调节作用。每日分别以生理盐水和低、中、高3个剂量的L–茶氨酸生理盐水溶液对D–半乳糖致衰老大鼠进行干预性处理8周后,检测大鼠的脾脏和胸腺器官指数、肝脏ALT和AST活力、肝脏内AGEs含量及炎性介质的mRNA表达水平和蛋白表达水平。结果显示:L–茶氨酸能提高D–半乳糖致衰老大鼠脾脏指数和胸腺指数,抑制肝脏ALT和AST活力上调,降低AGEs含量,并同时抑制肿瘤坏死因子–α(TNF–α)、肿瘤坏死因子受体–1(TNFR1)、白介素–1β(IL–1β)、白介素–1受体(IL–1R)、白介素6(IL–6)、诱导型一氧化氮合酶(iNOS)、细胞间粘附分子–1(ICAM–1)和血管细胞粘附分子–1(VCAM–1)等炎性介质的mRNA水平和蛋白水平,维持肝脏内环境稳定,其中,L–茶氨酸中、高剂量组对D–半乳糖致衰老大鼠肝脏保护作用较好。可见,L–茶氨酸可以通过提高机体免疫力,降低肝脏AGEs和炎性介质水平,维持肝脏内环境稳态,具有干预肝脏炎性衰老的功效。

      Thirty SD rats were randomly divided into five groups: normal group, model group and low, medium and high dose L-theanine group according to their body weight. The aging rat model was induced by daily subcutaneous injection of 200 mg/kg d-galactose physiological saline solution to investigate the regulation of L-theanine on the activity of alanine transaminase(ALT) and aspartate transaminase(AST), the contents of advanced glycation end products(AGEs) and inflammatory mediators in the liver. D-galactose-induced aging rats were treated with physiological saline, low, medium and high doses(100, 200, 400 mg/kg) of L-theanine physiological saline solution for 8 weeks. The organ index of spleen and thymus, the activity of ALT and AST, the contents of AGEs and the mRNA and protein levels of inflammatory mediators were measured after the treatment. The results showed that L-theanine could increase the spleen index and thymus index of d-galactose-induced aging rats, inhibit the up-regulation of liver ALT and AST activity, down-regulate the content of AGEs in the liver, and at the same time inhibit mRNA levels and protein levels of inflammatory mediators such as tumor necrosis factor-α(TNF-α), tumor necrosis factor receptor-1(TNFR1), interleukin 1 beta(IL-1β), interleukin-1 receptor(IL-1R), interleukin 6(IL-6), inducible nitric oxide synthase(iNOS), intercellular adhesion molecule -1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1), to maintain inflammatory internal environment stability in the liver. Among them, L-theanine in the middle and high dose groups had a better protective effect on liver of d-galactose-induced rats. In summary, L-theanine can improve immunity, inhibit the level of liver AGEs and inflammatory mediators, maintain homeostasis in the liver, and have the potential to prevent liver inflamm-aging.
